It enables directional insertion of any amplified DNA product into any linearized vector at any site. Firstly, the vector is linearized at the cloning site. A small sequence overlapped with each end of the cloning site is added onto the insert through PCR. The insert and the linearized vector, with overlapped sequences of 15 bp-20 bp on both 5’- and 3’-end, respectively, are mixed and incubated with Exnase II at 37℃ for 30 min (Fig. 1.). The cloning products can then be directly transformed to competent cells with a ture positive rate > 95%.
ClonExpress Entry One Step Cloning Kit is specially designed for entry cloning based on ClonExpress One Step Cloning technology. The primers of insert should contain 15 bp of constant sequence at 5’-ends homologous to the cloning site of CE Entry Vector provided in this kit. Mix the PCR products of insert and the CE Entry Vector, and incubate for 30 min. Your insert will rapidly and accurately fuse into the vector. During reaction, the homologous recombinase Exnase Entry can catalyze efficiently circularization of amplified insert and CE Entry Vector (up to 100 cfu/ng Vector), with a ture positive rate higher than 95%. In addition, Exnase Entry is compatible with most PCR reaction systems, which means that PCR products can be directly used for recombination without further purification. Compared with TA cloning, the ClonExpress Entry cloning approach is more compatible, more efficient, easier, and is capable of cloning longer fragments.
